dc.description.abstract This doctoral thesis presents a highly integrated multimodal neuroimaging investigation of the dopaminergic nigrostriatal system in rats, combining optogenetic stimulation of the substantia nigra (SN) with simultaneous PET and BOLD-fMRI. The work consists of three interconnected parts that collectively redefine how we interpret and methodologically approach brain connectivity and dopamine signaling in vivo. The first part established a pipeline for concurrent preclinical fPET/fMRI connectivity analysis. We demonstrated that metabolic connectivity derived from [18F]FDG-PET during optogenetic SN stimulation was a significantly more reliable marker of directed anatomical projection pathways than conventional BOLD-based hemodynamic connectivity. While hemodynamic connectivity was highly variable, seed- dependent, and often dissociated the stimulated SN from its known targets, metabolic connectivity faithfully recapitulated monosynaptic anterograde circuits. The second part investigated the influence of standard small-animal fMRI preparation protocols on [11C]raclopride PET tracer kinetics. This work evaluated the suitability of different anesthesia protocols—specifically isoflurane, α-chloralose, and a combination of isoflurane/medetomidine—for simultaneous PET/fMRI studies. The findings demonstrated that mechanical ventilation delayed tracer delivery, while the choice of anesthetic agent significantly influenced both tracer delivery and washout. Building on these foundations, the third part achieved the first simultaneous, time- resolved mapping of optogenetically evoked phasic dopamine release and its downstream hemodynamic consequences in the intact rat brain. Strikingly, robust dopamine release in the striatum and nucleus accumbens coincided with sustained negative BOLD responses in the same regions. While these findings demonstrated a clear association between phasic dopamine release and negative BOLD signal changes, the underlying mechanism likely involves a combination of factors, including GABA co-release, the direct vasoactive effects of dopamine, or a decoupling of the BOLD signal from underlying oxygen metabolism. These results suggest that in dopaminergic projection areas, neuronal activation can be paradoxically associated with negative BOLD signal changes. en

dc.description.abstract Diese Dissertation präsentiert eine hochintegrierte multimodale Neurobildgebungsstudie des dopaminergen nigrostriatalen Systems bei Ratten, die eine optogenetische Stimulation der Substantia nigra (SN) mit simultaner PET- und BOLD- fMRT-Bildgebung kombiniert. Die Arbeit gliedert sich in drei miteinander verknüpfte Teile, die gemeinsam einen erweiterten konzeptionellen und methodischen Rahmen für die Interpretation von Hirnkonnektivität und Dopamin-Signalübertragung in vivo schaffen. Im ersten Teil wurde eine Pipeline für die simultane präklinische fPET/fMRT- Konnektivitätsanalyse etabliert. Wir konnten zeigen, dass die aus der [¹⁸F]FDG-PET abgeleitete metabolische Konnektivität während der optogenetischen SN-Stimulation ein signifikant zuverlässigerer Marker für gerichtete anatomische Projektionsbahnen war, als die konventionelle, BOLD-basierte, hämodynamische Konnektivität. Während die hämodynamische Konnektivität hochgradig variabel und Seed-abhängig war und die stimulierte SN oft von ihren bekannten Zielregionen dissoziierte, bildete die metabolische Konnektivität monosynaptische, anterograde Netzwerke originalgetreu ab. Im zweiten Teil wurde der Einfluss von Standard-Präparationsprotokollen der Kleintier-fMRT auf die [¹¹C]Racloprid-PET-Tracerkinetik untersucht. Diese Arbeit bewertet die Eignung verschiedener Anästhesieprotokolle – insbesondere Isofluran, α- Chloralose und eine Kombination aus Isofluran und Medetomidin – für simultane PET/fMRT-Studien. Die Ergebnisse belegen, dass eine mechanische Beatmung die Tracer-Anflutung verzögert, während die Wahl des Anästhetikums sowohl die Anflutung als auch die Auswaschung des Tracers signifikant beeinflusst. Aufbauend auf diesen Grundlagen gelang im dritten Teil erstmals eine simultane, zeitlich aufgelöste Kartierung der optogenetisch ausgelösten phasischen Dopaminfreisetzung, sowie der daraus resultierenden hämodynamischen Veränderungen im intakten Rattengehirn. Bemerkenswerterweise fällt eine robuste Dopaminfreisetzung im Striatum und Nucleus accumbens mit anhaltenden negativen BOLD-Antworten in denselben Regionen zusammen. Obwohl diese Befunde auf eine klare Assoziation zwischen phasischer Dopaminfreisetzung und negativen BOLD-Signaländerungen hinweisen, spricht vieles dafür, dass der zugrundeliegende Mechanismus multifaktoriell ist und GABA-Cofreisetzung, direkte vasoaktive Effekte von Dopamin sowie eine Entkopplung zwischen BOLD-Signal und Sauerstoffmetabolismus umfasst. Die Ergebnisse deuten darauf hin, dass neuronale Aktivierung in dopaminergen Projektionsgebieten paradoxerweise mit negativen BOLD-Signaländerungen einhergehen kann. de_DE
