<title>Multimodal Imaging of Nigral Modulation: An Optogenetic PET/fMRI Study in Rats</title>

<description>Multimodal Imaging of Nigral Modulation: An Optogenetic PET/fMRI Study in Rats

This doctoral thesis presents a highly integrated multimodal neuroimaging&#13;
investigation of the dopaminergic nigrostriatal system in rats, combining optogenetic&#13;
stimulation of the substantia nigra (SN) with simultaneous PET and BOLD-fMRI. The&#13;
work consists of three interconnected parts that collectively redefine how we interpret&#13;
and methodologically approach brain connectivity and dopamine signaling in vivo.&#13;
The first part established a pipeline for concurrent preclinical fPET/fMRI&#13;
connectivity analysis. We demonstrated that metabolic connectivity derived from&#13;
[18F]FDG-PET during optogenetic SN stimulation was a significantly more reliable&#13;
marker of directed anatomical projection pathways than conventional BOLD-based&#13;
hemodynamic connectivity. While hemodynamic connectivity was highly variable, seed-&#13;
dependent, and often dissociated the stimulated SN from its known targets, metabolic&#13;
connectivity faithfully recapitulated monosynaptic anterograde circuits.&#13;
The second part investigated the influence of standard small-animal fMRI&#13;
preparation protocols on [11C]raclopride PET tracer kinetics. This work evaluated the&#13;
suitability of different anesthesia protocols—specifically isoflurane, α-chloralose, and a&#13;
combination of isoflurane/medetomidine—for simultaneous PET/fMRI studies. The&#13;
findings demonstrated that mechanical ventilation delayed tracer delivery, while the&#13;
choice of anesthetic agent significantly influenced both tracer delivery and washout.&#13;
Building on these foundations, the third part achieved the first simultaneous, time-&#13;
resolved mapping of optogenetically evoked phasic dopamine release and its downstream&#13;
hemodynamic consequences in the intact rat brain. Strikingly, robust dopamine release in&#13;
the striatum and nucleus accumbens coincided with sustained negative BOLD responses&#13;
in the same regions. While these findings demonstrated a clear association between phasic&#13;
dopamine release and negative BOLD signal changes, the underlying mechanism likely&#13;
involves a combination of factors, including GABA co-release, the direct vasoactive&#13;
effects of dopamine, or a decoupling of the BOLD signal from underlying oxygen&#13;
metabolism. These results suggest that in dopaminergic projection areas, neuronal&#13;

fMRT-Bildgebung kombiniert. Die Arbeit gliedert sich in drei miteinander verknüpfte&#13;
Teile, die gemeinsam einen erweiterten konzeptionellen und methodischen Rahmen für&#13;
die Interpretation von Hirnkonnektivität und Dopamin-Signalübertragung in vivo&#13;
schaffen.&#13;
Im ersten Teil wurde eine Pipeline für die simultane präklinische fPET/fMRT-&#13;
Konnektivitätsanalyse etabliert. Wir konnten zeigen, dass die aus der [¹⁸F]FDG-PET&#13;
abgeleitete metabolische Konnektivität während der optogenetischen SN-Stimulation ein&#13;
signifikant zuverlässigerer Marker für gerichtete anatomische Projektionsbahnen war, als&#13;
die konventionelle, BOLD-basierte, hämodynamische Konnektivität. Während die&#13;
hämodynamische Konnektivität hochgradig variabel und Seed-abhängig war und die&#13;
stimulierte SN oft von ihren bekannten Zielregionen dissoziierte, bildete die metabolische&#13;
Konnektivität monosynaptische, anterograde Netzwerke originalgetreu ab.&#13;
Im zweiten Teil wurde der Einfluss von Standard-Präparationsprotokollen der&#13;
Kleintier-fMRT auf die [¹¹C]Racloprid-PET-Tracerkinetik untersucht. Diese Arbeit&#13;
bewertet die Eignung verschiedener Anästhesieprotokolle – insbesondere Isofluran, α-&#13;
Chloralose und eine Kombination aus Isofluran und Medetomidin – für simultane&#13;
PET/fMRT-Studien. Die Ergebnisse belegen, dass eine mechanische Beatmung die&#13;
Tracer-Anflutung verzögert, während die Wahl des Anästhetikums sowohl die Anflutung&#13;
als auch die Auswaschung des Tracers signifikant beeinflusst.&#13;
Aufbauend auf diesen Grundlagen gelang im dritten Teil erstmals eine simultane,&#13;
zeitlich aufgelöste Kartierung der optogenetisch ausgelösten phasischen&#13;
Dopaminfreisetzung, sowie der daraus resultierenden hämodynamischen Veränderungen&#13;
im intakten Rattengehirn. Bemerkenswerterweise fällt eine robuste Dopaminfreisetzung&#13;
im Striatum und Nucleus accumbens mit anhaltenden negativen BOLD-Antworten in&#13;
denselben Regionen zusammen. Obwohl diese Befunde auf eine klare Assoziation&#13;
zwischen phasischer Dopaminfreisetzung und negativen BOLD-Signaländerungen&#13;
hinweisen, spricht vieles dafür, dass der zugrundeliegende Mechanismus multifaktoriell&#13;
ist und GABA-Cofreisetzung, direkte vasoaktive Effekte von Dopamin sowie eine&#13;
Entkopplung zwischen BOLD-Signal und Sauerstoffmetabolismus umfasst. Die&#13;
Ergebnisse deuten darauf hin, dass neuronale Aktivierung in dopaminergen&#13;
Projektionsgebieten paradoxerweise mit negativen BOLD-Signaländerungen&#13;
einhergehen kann.

<title>Vergleich der Prävalenzen asymptomatischer Low-Grade-Infekte im Implantatlager nach Entfernung von Osteosynthesematerial und Prothesenkomponenten bei Wechseloperationen. Eine prospektive Studie.</title>

Bei Prothesenwechseln nach endoprothetischem Gelenkersatz und bei&#13;
Metallentfernung nach Osteosynthesen zur Frakturversorgung werden trotz&#13;
fehlender Infektionzeichen in der Literatur Low-Grade-Infektionen in 17-40%&#13;
beschrieben (Dapunt et al., 2015; Fuchs et al., 2018). Zum Nachweis können&#13;
Abstriche, Gewebeproben oder histologische Gewebeuntersuchungen&#13;
angewendet werden.&#13;
In dieser Studie sollte überprüft werden, wie hoch die Raten von Low-GradeInfekten bei PatientInnen aus der Orthopädie und Unfallchirurgie bei Prothesenwechsel und Metallentfernung sind und welche prädisponierenden&#13;
Risikofaktoren dabei bedeutsam sein können. Es sollte geklärt werden, welche&#13;
diagnostischen Untersuchungen zur Feststellung eines Low-Grade-Infektes am&#13;
besten geeignet sind und welche Konsequenzen für die Therapie daraus&#13;
gezogen werden können. Dazu wurden prospektiv alle bis dahin&#13;
asymptomatischen PatientInnen mit Prothesenwechseln und Metallentfernungen&#13;
vom 01.06.2023 bis zum 31.12.2024 in die Studie aufgenommen. 369&#13;
PatientInnen wurden eingeschlossen, davon 262 (71%) Metallentfernungen und&#13;
107 Wechseloperationen (29%).&#13;
Intraoperativ wurden aus dem Implantatlager Abstriche, Gewebeproben zur&#13;
mikrobiologischen Untersuchung und ein Gewebestreifen zur histologischen&#13;
Untersuchung entnommen. Die Gewebestreifen wurden nach HE-Färbung nach&#13;
der Focus Klassifikation bezüglich der Anzahl der nachgewiesenen neutrophilen&#13;
Granulozyten eingeteilt. Es wurden die Daten der PatientInnen wie Alter,&#13;
Geschlecht, BMI, die Art und Lokalisation der eingebrachten Implantate, sowie&#13;
die präoperativen Entzündungsparameter (Leukozyten und CRP), als auch die&#13;
Nierenfunktionswerte (Harnstoff und Kreatinin) dokumentiert. Ebenso wurde&#13;
auch die Qualifikation des Operateurs bzw. der Operateurin bei der Erstoperation&#13;
erfasst. Die Behandlungsverläufe der PatientInnen mit Low-Grade-Infekten&#13;
wurden im Rahmen einer Nachuntersuchung nach durchschnittlich 13,7 Monaten&#13;
erhoben.&#13;
Die Ergebnisse zeigen hochsignifikante Unterschiede in der Infektionsrate&#13;
zwischen Orthopädie und Unfallchirurgie, insbesondere mit einer höheren&#13;
Infektionsrate in der Unfallchirurgie (30 % vs. 2 %). Signifikante Risikofaktoren&#13;
für Infektionen waren das männliche Geschlecht und Plattenosteosynthesen bei&#13;
Klavikula- und Oberarmkopffrakturen. Low-Grade-Infektionen wurden signifikant&#13;
häufiger festgestellt, wenn im präoperativen Labor vor Prothesenwechsel oder&#13;
Metallentfernung erhöhte Leukozyten- und CRP-Werte gemessen wurden.&#13;
Cutibacterium acnes war der am häufigsten nachgewiesene Verursacher von&#13;
Low-Grade Infekten. Gewebeproben hatten mit einer Sensitivität von 76% und einer Spezifität von 98% eine höhere Prädiktion für Low-Grade-Infektionen als Abstriche (Sensitivität 48% und Spezifität 99%) und histologische Untersuchungen (Sensitivität 31%&#13;
und Spezifität 99%). In fast der Hälfte der Low-Grade-Infektionen passte die&#13;
perioperative Antibiotikaprophylaxe nicht zum nachgewiesenen Keim. Auf&#13;
Grundlage der aktualisierten AWMF S3 Leitlinie vom Dezember 2024 sollen bei&#13;
Osteosynthesen und Prothesen an der Schulterregion die perioperative&#13;
Antibiotikaprophylaxe von Cefazolin auf Ampicillin/Sulbactam (Unacid©)&#13;
angepasst werden. Entsprechend der vorliegenden Studienergebnisse und der&#13;
Leitlinienempfehlungen wurden die klinikinternen SOPs im Hinblick auf&#13;
zukünftige Behandlungen dieser Art ebenfalls angepasst. Nun soll auch die&#13;
routinemäßige Entfernung der eingebrachten Plattenosteosynthesen an&#13;
Klavikula und Oberarmkopf nach abgeschlossenem Heilungsverlauf auch bei&#13;
asymptomatischen PatientInnen erfolgen. Durch dieses Vorgehen soll die Rate&#13;
der Low-Grade-Infekte verringert werden. In keinen der nachfolgenden Follow&#13;
ups zeigten sich weitere klinische Auffälligkeiten.

<title>Neural Dynamics of Numerical Cognition: Coding, Maintenance, and Interference in the Primate Frontoparietal Network</title>

This dissertation investigates how the brain of non-human primates encodes, retains&#13;
and recognizes numerical information, combining behavioral and neuronal approaches.&#13;
Understanding how numerical cognition emerges from distributed neural processing&#13;
offers a unique opportunity to explore the interaction between working memory,&#13;
recognition memory and decision-making – three core components of higher cognition. &#13;
&#13;
Electrophysiological recordings from the dorsolateral prefrontal (dlPFC) and posterior&#13;
parietal (PPC) cortices of rhesus monkeys performing a sequential delayed match-to-numerosity task revealed that single neurons encode recognition memory for numerical&#13;
quantities. During comparisons of the sample stimulus with up to three test stimuli, the&#13;
neurons exhibited distinct yet interacting patterns of familiarity modulation, including&#13;
repetition suppression and match enhancement. These results suggest that recognition&#13;
memory encompasses abstract, non-symbolic representations, and that the ability to&#13;
detect familiarity is distributed across prefrontal and parietal networks.&#13;
 &#13;
At the population level, the interaction between dlPFC and PPC was further&#13;
characterized by analyzing simultaneously recorded neuronal activity using canonical&#13;
correlation analysis (CCA). This approach revealed dynamic, direction-specific&#13;
coupling between both regions that varied across task phases. During stimulus&#13;
encoding, information flow was predominantly feedforward from parietal to prefrontal&#13;
areas, whereas maintenance periods were marked by more balanced interactions.&#13;
These results suggest that numerical recognition arises from the flexible coordination&#13;
of the frontoparietal network as a whole, rather than from the isolated processing of a&#13;
single brain region.&#13;

Extending these findings to a comparative perspective, behavioral performance in the&#13;
same task was examined across species, revealing that rhesus monkeys and carrion&#13;
crows show comparable accuracy and numerical effects, such as distance and size&#13;
effects. Despite these similarities, the temporal dynamics of decision-making differed&#13;
systematically between species. Monkeys exhibited increasing reaction times across&#13;
successive test phases, whereas crows responded progressively faster. Machinelearning&#13;
analyses further demonstrated that reaction times carried information about numerosity and task phase in monkeys but not in crows, suggesting species-specific&#13;
strategies in numerical recognition and decision control.&#13;
 &#13;
Together, these studies provide a comprehensive, multi-level account of numerical&#13;
cognition, covering from single-neuron encoding and network dynamics to behavior&#13;
across species. By integrating neurophysiological and behavioral evidence, this thesis&#13;
sheds light on abstract cognitive functions, such as number recognition.

In the last two decades, amid EU enlargement and the Eurozone crisis, many European coun-tries have experienced large-scale population loss through intra-EU mobility, fueling skill and labour shortages, population ageing and democratic backsliding. Yet emigration has attracted far less scholarly attention than immigration, and its specific political consequences for govern-ing parties in origin states remain poorly understood. This dissertation addresses that gap by asking: how do governing parties in EU origin states navigate a problem they cannot solve?&#13;
The dissertation argues that emigration constitutes a structurally blame-laden issue: it is highly visible, readily attributed to domestic political and economic conditions, and nearly impossible to address through available policy instruments. Two constraints are central. Legally, EU free movement law makes it impossible for national governments to restrict exit or force emigrants to return. Structurally, the main drivers of emigration – e.g., persistent wage gaps, labour market dysfunction, and migration networks – cannot be reversed within a single legislative term.&#13;
Three empirical articles examine governing parties’ responses to emigration at successive lev-els. The first (“Emigration – Who Cares?”), drawing on 938 party manifestos across all 27 EU member states (2004–2023), maps how, and under what conditions and by whom emigration is politicised. The second (“Please come back!”) analyses an original dataset of 81 national-level emigrant return policies across 17 member states (2004–2023) and gives insights into why gov-ernments adopt such programmes despite their limited effectiveness. The third (“Who Gets to Return?”) uses the same dataset alongside case studies of Ireland and Portugal to show how the universalist rhetoric of return policies masks systematic, party-family-specific selectivity in who actually receives meaningful return incentives. Taken together, the three articles point to a dual political logic of blame avoidance and distributive politics: blame avoidance explains the framing of emigration and the need for visible action, while distributive politics explains why that action takes partisan and selective forms.; Die Dissertation ist gesperrt bis zum 02.
